چکیده

Type XVII collagen (BP180), promotes adhesion of basal keratinocytes to epidermal basement membrane, and is the target of disease in patients with certain inherited or acquired blistering diseases. Therefore, the aim of the present study was to investigate IgA autoantibodies using recombinant BP 180 proteins against bullous pemphigoid in the sera of patients. Thirty consecutive BP sera were analyzed by ELISA and western blotting using highly purified recombinant subdomain forms of the collagen XVII proteins. We have recombinantly produced of the collagen XVII ecto-subdomains in a mammalian expression system (BP180-A, B, C and D). The patients Sera with bullous pemphigoid mainly react with epitopes clustered within the NC16A and the NC1, C-terminal domains of BP180. Interestingly, out of 30 patients with BP, the sera of 25 reacted with recombinant BP 180 (A, B and C), and the sera of 22 reacted with BP 180-D. The bullous pemphigoid patient’s sera reacted with the recombinant BP180 ectodomain (A, C and D). Therefore, a sensitivity of 86. % with BP180 (A and C) and 73% with BP180-D, resulted for the ELISA. Our results demonstrate that autoantibodies in BP target epitopes of subdomains of BP180 ectodomain.

Development of an ELISA for sensitive and specific detection of IgA autoantibodies against BP180 in pemphigoid diseases

BACKGROUND Pemphigoids are rare diseases associated with IgG, IgE and IgA autoantibodies against collagen XVII/BP180. An entity of the pemphigoid group is the lamina lucida-type of linear IgA disease (IgA pemphigoid) characterized by IgA autoantibodies against BP180. While for the detection of IgG and IgE autoantibodies specific to collagen XVII several ELISA systems have been established, no q...

Autoantibodies to bullous pemphigoid antigen 180 induce dermal-epidermal separation in cryosections of human skin.

Bullous pemphigoid is a subepidermal autoimmune blistering disease associated with autoantibodies to the hemidesmosomal bullous pemphigoid antigens 180 and 230. Most sera from bullous pemphigoid patients recognize epitopes within the N-terminal NC16A portion of the bullous pemphigoid 180 ectodomain. Using cryosections of human skin, patients' sera were shown to generate dermal-epidermal separat...

BP180 as the common autoantigen in blistering diseases with different clinical phenotypes.

Bullous pemphigoid antigen 180 (BP180, type XVII collagen) is a transmembrane hemidesmosomal glycoprotein of basal keratinocytes that spans the lamina lucida of the dermal-epidermal junction. Five autoimmune subepidermal blistering diseases are associated with an immune response to BP180, including bullous pemphigoid (BP), pemphigoid gestations (PG), cicatricial pemphigoid (CP), lichen planus p...

A Case of Chronic Bullous Disease of Childhood That Was Reactive to the Antigen of 120 kDa (LAD-1).

Chronic bullous disease of childhood (CBDC) is an autoimmune blistering disease that is characterized by Immunoglobulin A (IgA) deposits at the basement membrane zone. IgA autoantibodies (aAbs) from the serum of patients with CBDC react with antigens of 97 kDa (LABD97) and 120 kDa (LAD-1), and both of which are fragments of the extracellular domain of bullous pemphigoid 180 (BP180, type XVII co...
